ABSTRACT

Abstract: Climatic variation throughout the day influences the ecophysiological performance of plants at different growth stages and phases. Therefore, this work aimed to evaluate the effect of climatic variation on ecophysiological aspects of T. roseoalba and H. heptaphyllus at different hours of the day and indicate the ideal time for measuring ecophysiological variables in these species. The research was carried out in a greenhouse at the forest nursery of the Federal University of Paraíba, Campus II, in the municipality of Areia, Paraíba state, Northeastern Brazil. The experimental design was completely randomized, consisting of 10 evaluation times throughout the day (from 8 am to 5 pm), with 1 h hour interval between each evaluation. Temperature and air relative humidity inside and outside the greenhouse were evaluated to understand the effect on gas exchange (net assimilation rate of CO2, stomatal conductance, transpiration rate, internal concentration of CO2, and vapor-pressure deficit) and chlorophyll a fluorescence (initial, maximum, and variable fluorescence, photochemical quenching, and electron transport rate). Data were submitted to canonical correlation analysis and principal component analysis to verify the relationship between climatic and ecophysiological variables. For both species, higher correlation was found between internal and external relative humidity with all the ecophysiological variables analyzed, except for initial fluorescence. Thus, climatic factors influenced the photosynthetic performance of T. roseoalba and H. heptaphyllus plants, and 8 am to 9 am is indicated for carrying out ecophysiological evaluations in both species.

ABSTRACT

H. impetiginosus belongs to the Bignoniaceae family; it has a great potential for economic exploitation and can be used in landscaping of urban areas, reforestation, recovery of degraded areas, and folk medicine. The experiment was carried out to evaluate the effect of light and temperature regimes on the germination and vigor of Handroanthus impetiginosus seeds at the Seed Analysis Laboratory of UFRPE/UAG. The seeds were subjected to light regimes: white, far red, red, and no light at 15°C, 20°C, 25°C, 30°C, 35°C, and 40°C, using a completely randomized experimental design in a factorial scheme (4 × 6), with four repetitions of 25 seeds. The different light regimes did not influence the seed germination of H. impetiginosus. The highest germination percentage (92%) and germination speed index (7.94) were obtained at temperatures 28.2°C and 29.2°C, respectively, both under red light. The longest seedling length was also obtained from the seeds subjected to red light regime at 25°C. The temperatures of 15°C and 40°C inhibited the germination of H. impetiginosus seeds. H. impetiginosus seeds are classified as neutral photoblastics, and constant temperatures of 28.2°C and 29.2°C provided maximum germination.


H. impetiginosus, pertencente à família Bignoniaceae, apresenta grande potencial para exploração econômica, podendo ser utilizado no paisagismo de áreas urbanas, reflorestamentos, recuperação de áreas degradadas e na medicina popular. O experimento foi conduzido no Laboratório de Análise de Sementes da UFRPE/UAG com o objetivo de avaliar o efeito dos regimes de luz e temperatura na germinação e vigor de sementes de Handroanthus impetiginosus. As sementes foram submetidas aos regimes de luz: branca, vermelho distante, vermelho e ausência de luz sob as temperaturas de 15, 20, 25, 30, 35 e 40°C, sendo utilizado o delineamento experimental inteiramente ao acaso, em esquema fatorial (4x6), com quatro repetições de 25 sementes. Os diferentes regimes de luz não influenciaram na germinação de sementes de H.impetiginosus. A maior porcentagem de germinação (92%) e índice de velocidade de germinação (7,94) foram obtidos nas temperaturas 28,2 e 29,2°C, respectivamente, ambos no regime de luz vermelha. O maior comprimento de plântula também foi obtido de sementes submetidas ao regime de luz vermelha na temperatura de 25ºC. As temperaturas de 15ºC e 40ºC inibiram a germinação das sementes de H. impetiginosus. As sementes de H. impetiginosus são classificadas como fotoblásticas neutras e as temperaturas constantes de 28,2 e 29,2°C proporcionaram máxima germinação.

Resumen: Introducción: Las enfermedades pulmonares intersticiales son un grupo muy heterogéneo de afecciones que tienen manifestaciones clínicas, radiológicas y funcionales en común. Su epidemiología está documentada en forma parcial y su etiología es muy variada. El diagnóstico se basa en la clínica, la tomografía computada de alta resolución y, en algunos casos, la histología. Objetivos: describir la población de pacientes con diagnóstico de enfermedad pulmonar intersticial asistidos en la Policlínica de Intersticiopatías del Hospital Pasteur en el período comprendido entre el 5 de mayo de 2017 y el 17 de mayo de 2019. Materiales y métodos: Estudio descriptivo, observacional y transversal. Se utilizó un muestreo por conveniencia. Las variables analizados fueron: edad, sexo, comorbilidades, etiología, patrón tomográfico predominante, funcional respiratorio (Test de Marcha 6 Minutos y espirometría forzada con difusión de monóxido) y tratamiento instituido. Se obtuvo consentimiento informado y el trabajo fue aprobado por el Comité de Bioética del Hospital Pasteur. Resultados: 20 de los 42 pacientes asistidos eran portadores de enfermedad pulmonar intersticial. Predominó el sexo femenino (14 pacientes, 70%). La edad media fue 60 ± 16.84 años. La comorbilidad más frecuente fue el tabaquismo (13 pacientes, 65%). En 6 pacientes (30%) se identificaron exposiciones ambientales o laborales de riesgo. La etiología fue determinada en 15 pacientes (75%), siendo en 10 pacientes (50%) asociada a enfermedades autoinmunes sistémicas y en 3 pacientes (15%) neumonitis por hipersensibilidad. La mayoría (11 pacientes, 55%) tuvieron patrones tomográficos inespecíficos. 94.11% tuvieron un patrón restrictivo en el estudio funcional respiratorio, predominando la restricción moderada a severa. La difusión monóxido media fue 47.25 ± 16.71%. 11 pacientes (55%) fueron tratados con corticoides sistémicos y 2 pacientes necesitaron oxígeno a permanencia. Conclusiones: Las características demográficas, etiología, patrones tomográficos y tratamiento instituidos están influenciados por un sesgo de selección, ya que una elevada proporción de pacientes son derivados desde policlínicas de enfermedades autoinmunes sistémicas. Es fundamental un manejo integral de estos pacientes porque la asociación con otras enfermedades que pueden contribuir en la sintomatología es frecuente. Las enfermedades pulmonares intersticiales encontradas son mayormente severas y de diagnóstico tardío.


Abstract: Introduction: Interstitial lung diseases are a very heterogeneous group of conditions that have common clinical, radiological and functional manifestations. Its epidemiology is partially documented and its etiology is very varied. The diagnosis is based on the clinic, high resolution computed tomography and, in some cases, histology. Objectives: to describe the population of patients diagnosed with interstitial lung disease assisted at the Hospital Pasteur Polyclinic for Interstitiopathies in the period between May 5, 2017 and May 17, 2019. Materials and methods: Descriptive, observational and cross. Convenience sampling was used. The variables analyzed were: age, sex, comorbidities, etiology, predominant tomographic pattern, respiratory function (6-minute walk test and forced spirometry with monoxide diffusion) and instituted treatment. Informed consent was obtained and the work was approved by the Bioethics Committee of the Pasteur Hospital. Results: 20 of the 42 assisted patients were carriers of interstitial lung disease. Female sex predominated (14 patients, 70%). The mean age was 60 ± 16.84 years. The most frequent comorbidity was smoking (13 patients, 65%). Environmental or occupational risk exposures were identified in 6 patients (30%). The etiology was determined in 15 patients (75%), being associated with systemic autoimmune diseases in 10 patients (50%) and hypersensitivity pneumonitis in 3 patients (15%). Most (11 patients, 55%) had nonspecific tomographic patterns. 94.11% had a restrictive pattern in the functional respiratory study, with moderate to severe restriction prevailing. The average monoxide diffusion was 47.25 ± 16.71%. 11 patients (55%) were treated with systemic corticosteroids and 2 patients required oxygen permanently. Conclusions: The demographic characteristics, etiology, tomographic patterns and instituted treatment are influenced by a selection bias, since a high proportion of patients are derived from polyclinics for systemic autoimmune diseases. Comprehensive management of these patients is essential because the association with other diseases that may contribute to the symptoms is frequent. The interstitial lung diseases found are mostly severe and late diagnosed.

A etiologia foi determinada em 15 pacientes (75%), sendo associada a doenças autoimunes sistêmicas em 10 pacientes (50%) e pneumonite por hipersensibilidade em 3 pacientes (15%). A maioria (11 pacientes, 55%) apresentava padrões tomográficos inespecíficos. 94,11% apresentaram padrão restritivo no estudo respiratório funcional, prevalecendo restrição moderada a grave. A difusão média de monóxido foi de 47,25 ± 16,71%. 11 pacientes (55%) foram tratados com corticosteróides sistêmicos e 2 pacientes necessitaram de oxigênio permanentemente. Conclusões: As características demográficas, etiologia, padrões tomográficos e tratamento instituído são influenciados por um viés de seleção, pois uma alta proporção de pacientes é derivada de policlínica para doenças autoimunes sistêmicas. O manejo abrangente desses pacientes é essencial, pois a associação com outras doenças que podem contribuir para os sintomas é frequente. As doenças pulmonares intersticiais encontradas são na maioria graves e diagnosticadas tardiamente.

ABSTRACT

@#The Philippine Interprofessional Education and Collaboration (PhIPEC) Conference is the first-ever national program held in the Philippines on interprofessional education and collaboration (IPEC). This project, initiated through an inter-university partnership between University of Santo Tomas and Angeles University Foundation, aimed to facilitate uniform understanding of IPEC across higher education institutions and health facilities as well as to instigate IPEC related researches in the country. The two-day conference was able to gather over 80 participants from more than 10 health and social care professions and 15 speakers who shared their expertise in health education and practice. Aside from these, the initiative has also gathered more than 500 followings in Facebook Page and 161 members in the mailing list. With the turn-out of this initiative, there was a move to rename the group into PhIPEC Network. The network has been agreed upon to serve as an informal entity that represents a collective of Filipino health and social care professionals towards advocating collaborative learning and health care services. Future directions were also determined focused on considering IPEC initiatives in education, practice, research, and policies.

ABSTRACT

Osteosarcoma is a malignant tumor of primitive bone cells with a high incidence in dogs and humans. The need for more effective drugs with less adverse consequences has pushed the development of chemotherapeutic agents from plants and other natural sources. The aim of this study was to verify the cytotoxic effects of beta-lapachone, a compound present in the sawdust of Tabebuia sp. (popularly known as ipê) wood, on canine osteosarcoma cells subcultured and treated in different concentrations (0.1µm, 0.3µm e 1.0µm) and exposure times (24h, 48h e 72h). Results were obtained through Trypan blue dye exclusion, tetrazolium reducing method, cell survival assay, Annexin V-FITC and Propidium Iodine labeling, JC-1 dye labeling and cell cycle kinetics e analysis. The group treated with 0.3µm beta-lapachone presented higher decrease in cell viability (80.27%, 24h, 47.41%, 48h and 35.19%, 72h) and greater progression of cytotoxicity (19.73%, 24h, 52.59%, 48h and 64.81%, 72h). The lower IC50 (0.180µm) was verified in the group treated for 72 hours. Cell growth after treatment decreased as concentration and time of exposure increased, with 0.50% survival fraction at the concentration of 1.0µm. Initial apoptosis was the most frequent type of cell death in all groups, reaching bottom in the 24-hour group treated with 0.1µm (4.26%) and peaking in the 72-hour group treated with 1.0µm (85.89%). Mitochondrial depolarization demonstrated a dose-dependent phenomenon, indicating the intrinsic apoptosis. Cell growth inhibition by blocking cell cycle in the G0/G1 phase related to the exposure the time. β-lapachone is cytotoxic for canine osteosarcoma cells, induces apoptosis and promotes cell cycle arrest in G0/G1 phase.(AU)


O osteossarcoma é o tumor maligno das células ósseas primitivas, com alta incidência em cães e humanos. A necessidade de medicamentos mais efetivos, com menor consequência adversa, tem gerado esforços para o desenvolvimento de agentes quimioterápicos compostos por plantas e outras fontes naturais. O objetivo deste estudo foi verificar os efeitos citotóxicos da beta lapachona, um composto presente na serragem da madeira do ipê, sobre células de osteossarcoma canino subcultivadas e submetidas ao tratamento, de acordo com as diferentes concentrações (0.1µm, 0.3µm e 1.0µm) e tempo de exposição (24h, 48h e 72h). Os resultados foram obtidos por meio dos métodos de exclusão do corante azul de Tripan e de redução do tetrazólio, além dos ensaios de sobrevivência celular, de dupla marcação com Anexina V-FITC e Iodeto de Propídio, de marcação com o corante JC-1 e pela análise da cinética do ciclo celular. O grupo tratado com 0.3µm de beta lapachona apresentou melhor regressão da viabilidade celular (80,27%, 24h; 47,41%, 48h e 35,19%, 72h) e maior progressão da citotoxicidade (19,73%, 24h; 52,59%, 48h e 64,81%, 72h). O menor IC50 (0.180µm) ocorreu no grupo tratado por 72 horas. O crescimento celular após o tratamento foi menor, de acordo com o aumento da concentração e tempo de exposição, apresentando 0,50% de fração de sobrevivência na concentração de 1.0µm. A apoptose inicial foi o tipo de morte celular mais frequente em todos os grupos, menor no grupo de 24 horas tratado com 0.1µm (4,26%) e maior no grupo de 72 horas tratado com 1.0µm (85,89%). A despolarização mitocondrial ocorreu de maneira dose dependente, indicando a ocorrência de apoptose intrínseca. A β lapachona possui efeitos citotóxicos em células de osteossarcoma canino, induz apoptose e promove o bloqueio do ciclo celular na fase G0/G1.(AU)

ABSTRACT

The purpose of this study was to investigate the immunomodulatory activity of ice plant (Mesembryanthemum crystallinum) extract (IPE) in vitro and in vivo. Raji (a human B cell line) and Jurkat (a human T cell line) cells were treated with various doses of IPE and cell proliferation was measured by WST assay. Results showed that IPE promoted the proliferation of both Raji and Jurkat cells in a dose-dependent manner. IPE also enhanced IL-6 and TNF-α production in macrophages in the presence of lipopolysaccharide (LPS), although IPE alone did not induce cytokine production. Moreover, IPE treatment upregulated iNOS gene expression in macrophages in a time- and dose-dependent manner and led to the production of nitric oxide in macrophages in the presence of IFNγ. In vivo studies revealed that oral administration of IPE for 2 weeks increased the differentiation of CD4+, CD8+, and CD19+ cells in splenocytes. These findings suggested that IPE has immunomodulatory effects and could be developed as an immunomodulatory supplement.

RESUMO A preocupação com o tratamento do Diabetes mellitus (DM) leva a uma crescente busca por terapias alternativas, como o uso de plantas medicinais, entre as quais, destaca-se o uso de Handroanthus heptaphyllus (Mart.) Mattos (popular Ipê roxo). Neste estudo realizamos a investigação química da presença de compostos fenólicos em H. heptaphyllus e o efeito do tratamento com o extrato aquoso da casca desta planta em parâmetros bioquímicos e nos níveis de lipoperoxidação tecidual e plasmática em animais diabéticos. Metodologia: Ratos Wistar machos foram submetidos ao desenvolvimento do quadro de DM por meio da administração intraperitoneal (IP) de Aloxano monohidrato (150 mg/Kg IP). Após a confirmação de hiperglicemia (>200 mg dL-1), os animais foram distribuídos nos grupos Diabético (D; n=6) e Diabético Tratado (DT; n=6). O tratamento consistiu na administração diária do extrato aquoso da casca de H. heptaphyllus via oral (v.o.) (150mg/Kg v.o.) por quatro semanas. O extrato aquoso foi analisado qualitativamente por cromatografia de camada delgada. Resultados: A análise qualitativa do extrato aquoso da casca indicou a presença de compostos fenólicos da subclasse flavonoides. O tratamento com o extrato aquoso reduziu a glicemia de jejum a partir da 3ª semana de tratamento, melhorou a resposta glicêmica à sobrecarga de glicose, diminuiu os níveis de triglicerídeos e índice LDL (Triglicerídeos/HDL). Estes resultados sugerem o uso terapêutico do extrato aquoso das cascas de H. heptaphyllus no tratamento do DM.


ABSTRACT Alternative medicine for diabetes mellitus (DM) treatment represents a growing research area on the use of medicinal plants, of which Handroanthus heptaphyllus (mart.) Mattos (popularly known as purple ipe) is most prominent. In this study, we investigated the presence of phenolic compounds and the effects of treatment with aqueous extract of in H. heptaphyllus in biochemical profile in plasma and the levels of lipid peroxidation in tissues and plasma in diabetic animals. Male Wistar rats were induced to develop DM through intraperitoneal (IP) administration of alloxan monohydrate (150 mg/kg IP). Once hyperglycemia (>200 mg dL-1) was confirmed, the animals were divided into the Diabetic (D; n=6) and Treated Diabetic (TD; n=6) groups. The TD group received daily administration (150 mg/kg v.o.) of aqueous extract of H. heptaphyllus for four weeks. The aqueous extract was also analyzed qualitatively by layer chromatography. Qualitative analysis of the aqueous extract of the bark indicated the presence of phenolic compounds from the flavonoid subclass. The treatment with the aqueous extract reduced fasting blood glucose levels from the third week of treatment on, improved the glycemic response to the glucose tolerance test, and lowered the levels of triglycerides and the LDL index (triglycerides/HDL). These findings suggest therapeutic use of the aqueous extract of H. heptaphyllus bark in treating DM.

Abstracts: Introduction: The concept of Interprofessional Education (IPE) in Indian health sciences education is lacking. The purpose of this study was to find out the perception of faculty from different health sciences faculties regarding IPE. Methodology: A modified version of the Readiness for Interprofessional Learning Scale (RIPLS) was used to find out the faculty perception regarding IPE. Results: 118 teachers from medical, physiotherapy and nursing responded to the survey. Majority of them did not have any experience of IPE. Conclusion: Teachers agreed that IPE will prepare students for collaboration in healthcare teams, and felt that teaching as well as learning in interprofessional groups is possible. Faculty training in IPE is required.

ABSTRACT

<p> The WHO reported the importance of IPE (Interprofessional Education) in order to implement team-based medicine smoothly. Some Japanese medical educational institutes presented their IPE programs, which involved real patients, but most IPE programs used scenario-based or standardized patients. Moreover, few reports showed IPE programs for diabetic patient education. We created a new program, called the "Diabetes education class IPE" . Participants were 2 medical, 4 nursing, 4 pharmacy, and 3 dietician students. The students experienced the planning and management of and reflection on diabetes education classes. As a result of reflection, the "Diabetes education class IPE" was viewed as a useful IPE program.</p>

Foi elaborada uma chave interativa para a identificação das 26 espécies da Aliança Tabebuia (Bignoniaceae) nativas do estado da Bahia, Brasil. A partir de uma página web, contendo caixas de seleção com caracteres botânicos, de marcação livre e não sequencial, vão se eliminando as espécies que não atendem aos critérios selecionados até a identificação completa do material. A chave está inserida em um site, contendo informações sobre Bignoniaceae, a Aliança Tabebuia e o estado da Bahia, bem como um glossário dos termos botânicos utilizados na chave e um banco de imagens. O sistema foi desenvolvido a partir das linguagens HTML e Javascript, é autoexecutável em CD-ROM e está disponível para download em: http://www.mendeley.com/profiles/alessandro-rapini/publications/ComputerProgram/. A utilização das chaves interativas facilita e dinamiza o processo de identificação dos táxons, contribuindo para a difusão do conhecimento biológico e elaboração de programas voltados ao reconhecimento e conservação da biota.


An interactive key to identify the 26 species of the Tabebuia Alliance (Bignoniaceae) from the state of Bahia, Brazil, was developed. From a web page containing check boxes with botanical characters, with free and not sequential marking, species which not match to the criteria are eliminated until the complete identification of the material. The key is hosted in a website, that also provides general information about the Bignoniaceae, the Tabebuia Alliance and the state of Bahia, as well as a glossary of the botanical terminology employed in the key and a bank of images. The system was created using HTML and Javascript languages. It runs from a CD-ROM and is available for download at: http://www.mendeley.com/profiles/alessandro-rapini/publications/ComputerProgram/. The use of interactive keys facilitates and streamlines the process of taxa identification, contributing to the dissemination of biological knowledge and preparation of programs to the recognition and conservation of biota.

ABSTRACT

The aim of this study was to evaluate the effect of Tecoma stans L. Juss. ex Kunth seeds mass on initial emergence, growth and, seedling development under different light conditions. The seeds were separated in four mass classes and sowed in four replicates of 24 seeds for each class, under full sun and canopy shade. Under sun environment was observed a greater percentage of emergence. Heavy seeds presented the greater percentage of emergence under both environments, but a greater rate was observed under canopy shade. One month after the start of experiments, the seedlings at the shade environment presented 100 percent of mortality. The growth and development seedlings under full sun were noticed for five months. In this period, only in the first three months was possible to observe the effects of Tecoma stans seeds mass on capacity of seedlings to acquire dry mass. The seedlings biomass partitions were similar among the tested mass class. The seedlings of smaller mass tended to a high specific leaf area in relation to the seedlings from large seeds, mainly in the first three months, resulting in a great acquisition of dry mass by these seedlings. In the fourth month, the specific leaf area did not present any tendency. Because the biggest seeds to give rise seedlings with best initial development than smallest seeds can be considered as species reproductive strategy. To produce seeds of different sizes also can be considered as way of species to spread in many microhabitats.


Este estudo teve como objetivo avaliar o efeito da massa das sementes de Tecoma stans L. Juss. ex Kunth na emergência, no crescimento e no desenvolvimento inicial de suas plântulas sob diferentes condições de luminosidade. As sementes foram separadas em quatro classes de massa e semeadas em quatro réplicas de 24 sementes para cada classe, no sol e na sombra. No ambiente de sol ocorreu a maior porcentagem de emergência. Dentre as classes de massa, sementes maiores apresentaram maior porcentagem de emergência em ambos os ambientes, porém a maior velocidade foi observada na sombra. Após um mês do início do experimento as plântulas da sombra apresentaram 100 por cento de mortalidade. O crescimento e desenvolvimento das plântulas no sol foram acompanhados mensalmente por um período de cinco meses. Neste período, foi possível observar os efeitos da massa das sementes de Tecoma stans no ganho de massa seca das plântulas nos três primeiros meses; a partir deste, a massa das sementes não teve efeito no ganho de matéria seca pelas plântulas. A partição da biomassa das plântulas não diferiu entre as classes de massa testadas. As plântulas de menor massa apresentaram uma tendência a uma maior área foliar específica em relação às plântulas originárias de sementes mais pesadas, principalmente nos três primeiros meses, resultando em um ganho maior de matéria seca das plântulas de menor classe de massa. Já no quarto mês, a área foliar específica não apresentou qualquer tendência. O fato de sementes maiores originarem plântulas com melhor desenvolvimento inicial quando comparadas com as sementes menores pode ser considerada como estratégia reprodutiva da espécie, assim como produzir sementes de diversos tamanhos pode ser considerado como uma maneira da espécie dispersar-se e colonizar um maior número de microambientes.

ABSTRACT

PURPOSE: To study the antitumor action of Tabebuia avellanedae in experimentally induced colon carcinogenesis by azoxymethane in mice. METHODS: Fifty (n=50) mice were divided into five groups: in group I azoxymethane (AOM) was administered, in Group II - β-lapachone, in group III - vehicle (diluent) and in group IV - vehicle + AOM and finally in group V - β-lapachone + AOM. RESULTS: It was observed the presence of aberrant crypt foci in all animals of groups I and IV, 50 percent in group II and 90 percent in group V. CONCLUSION: The β-lapachone extracted from the Tabebuia avellanedae showed no protective effect of lesions induced by azoxymethane in colon of mice.


OBJETIVO: Estudar a ação antitumoral da Tabebuia avellanedae (Ipê-Roxo) na carcinogênese colônica induzida experimentalmente pelo azoximetano em camundongos. MÉTODOS: Foram utilizados 50 camundongos divididos em 5 grupos: grupo I administrado Azoximetano (AOM); grupo II - β-lapachona; grupo III - veículo (diluente); grupo IV - veículo + AOM; e grupo V - β-lapachona + AOM. RESULTADOS: Observou-se presença de focos de criptas aberrantes em todos os animais dos grupos I e IV, 50 por cento no grupo II e 90 por cento no grupo V. CONCLUSÃO: A β-lapachona extraída da Tabebuia avellanedae não apresentou efeito protetor das lesões induzidas pelo azoximetano em cólon de camundongos.

ABSTRACT

OBJETIVO: Estudo morfológico do efeito da sulfadiazina de prata, extrato de ipê-roxo e extrato de barbatimão na cicatrização de feridas cutâneas. MÉTODOS: Utilizou-se 96 ratos Wistar. Todos foram submetidos à ligadura da veia femoral direita para produzir hipertensão venosa. Após 30 dias foi confeccionada a ferida cutânea. Dividiu-se os animais em quatro grupos. O grupo S recebeu aplicação tópica de sulfadiazina de prata; o grupo IR, extrato de ipê-roxo; o grupo B, extrato de barbatimão e o grupo C, aplicação de solução salina a 0,9 por cento, diariamente, nas feridas por um período de sete, 14 e 30 dias. A análise histológica avaliou: proliferação vascular, neutrófilos, linfócitos, fibroblastos, fibras colágenas e epitelização. RESULTADOS: Os achados macroscópicos mostraram epitelização completa aos 14 dias em todos os animais dos grupos S, IR e B. Na análise histológica aos 14 dias, apenas o grupo C ainda apresentava epitelização incompleta em seis animais; neste mesmo período houve diferença estatisticamente significativa entre o grupo controle e os demais grupos quanto ao processo inflamatório e neovascularização. Em relação à presença de fibroblastos e colágeno, houve diferença estatisticamente significativa entre o grupo controle e os demais grupos aos 30 dias. CONCLUSÃO: A análise dos resultados morfológicos permite inferir que o grupo S, IR e B foram favorecidos no processo de cicatrização das feridas cutâneas, quando comparados com o controle.


OBJECTIVE: Morphological study of effects of silver sulfadiazine, Tabebuia avellanedae (ipê-roxo) extract and Stryphnodendron adstringens (barbatimão) extract on cutaneous wound healing was done. METHODS: Ninety six Wistar rats were used. All animals underwent a femoral right vein ligation to induce a venous hypertension. Thirty days after the cutaneous wound was done, they were divided into groups of four animals. Group S received silver sulfadiazine topical application; group IR, Tabebuia avellanedae extract topical application; group B received Stryphnodendron adstringens extract topical application and group C received physiological solution topical application, every day for a period of seven, 14 and 30 days. Histology analyzed the presence vascular proliferation, neutrophil and lymphocite, fibroblast, collagen fiber and epithelization. RESULTS: The macroscopic analysis showed complete epithelization at 14 days in group S, IR e B. The histological data at 14 days of observation only group C still showed incomplete epithelization in six animals. At the same period there was a statistic significant difference between the control group and the others groups as inflammation process and neovascularization. About the presence fibroblasts and collagen, there was statistic significant difference between the control group and the others groups at the 30 days because at this period the control group still showed fibroblasts and collagen lower when compared to the others groups. CONCLUSION: The morphological analysis of the results permits to infer that the group S, IR and B had a better healing of skin wounds, when compared with the control.

ABSTRACT

As mudas propagadas por técnicas de micropropagação geralmente apresentam alterações significativas induzidas pelas condições in vitro, que diminuem a capacidade de sobrevivência após a transferência para o ambiente ex vitro, sendo fundamental aavaliação da mudança estrutural durante o processo de adaptação. Objetivou-se com este trabalho, identificar as diferenças anatômicasfoliares entre plantas de Tabebuia serratifolia (Vahl) Nich. (ipê amarelo), cultivadas in vitro, in vivo e durante a aclimatização. Foram utilizadas plântulas mantidas por 43 dias em sala de crescimento, com 20, 40, 60 e 80 dias de aclimatização e mudas de 90 diascultivadas em viveiro. As plântulas foram obtidas via cultura de embriões em meio MS e transplantadas para tubetes contendo plantmax®, para aclimatização em viveiro, sob 50% de sombreamento. Nas mesmas condições da aclimatização, foram produzidas asmudas in vivo. Cortes transversais e paradérmicos foram preparados de acordo com técnicas usuais em microtecnia vegetal. As plântulas in vitro apresentam os tecidos foliares pouco diferenciados e estômatos maiores e mais abertos, exigindo maiores cuidadosna etapa inicial de aclimatização. Aos 60 dias de aclimatização as novas folhas produzidas possuem alguns aspectos anatômicos que podem conferir maior eficiência fotossintética e maior capacidade de regulação hídrica das plantas.


The seedlings spread by micropropagation techniques, usually show significant alterations induced by the conditions in vitro, which decrease the survival capacity after the transfer to conditions ex vitro. The evaluation of the structural change during theacclimatization is necessary for the understanding of this adaptation process. The objective of this research was to identify the anatomical differences among the seedlings cultivated in vitro, in vivo and acclimatized plants of Tabebuia serratifolia (Vahl) Nich. (yellow ipe), species with great medicinal and ornamental interest. One used seedlings maintained for 43 days in growth chamber, with 20, 40, 60 and 80 days of acclimatization and seedlings after 90 days of cultivation in vivo. Seedlings were obtained from culture of embryos in half BAD and transplanted to tubes containing plantmax®, for aclimatização in nursery house under 50% of shad. Inthe same conditions of the acclimatization the seedlings in vivo were produced. Transversal and paradermal sections were preparedusing the usual microthecniques. The results showed that the seedlings in vitro have the leaf tissue little differed and the stomata are open and bigger, demanding greater cares in the initial stage of acclimatization. At 60 days of acclimatization the new leaves produced have some anatomical aspects that can originate higher photosynthetic efficiency and better water regulation capacity of the plants.

Um fator importante na determinação da sobrevivência de uma espécie, é sua adaptação à condição de alta ou baixa luminosidade. O objetivo deste estudo foi verificar o desenvolvimento inicial, teores de clorofila e alocação de biomassa em plantas de Tabebuia heptaphyilla (Vell.) Tol, em três condições de luminosidade. O experimento foi conduzido em delineamento de blocos casualizados, com três níveis de luminosidade (pleno sol, 50 por cento de luz e sombra natural) e com 15 repetições em cada tratamento, sendo cada planta considerada como uma repetição. Foram avaliados a altura e diâmetro aos 60, 82, 103, 124, 145 dias após a emergência das plântulas. Plantas expostas ao ambiente de sombra natural apresentaram menor desempenho vegetativo do que as demais condições de cultivo testadas (50 por cento e 100 por cento de luminosidade). A condição de 50 por cento de luminosidade pode ser recomendada para a formação de mudas, no entanto, essa prática também pode ser realizada a pleno sol.


Important factor in determining species survival, for example, its adaptability to high or low conditions of luminosity. The objective of this study was to verify the initial development, chlorophyll content, and biomass allocation in plants of Tabebuia heptaphyilla (vell.) tol., in three lighting conditions. The experiment was carried out in randomized complete blocks, with three levels of luminosity (full sunlight, 50 percent of light, and natural shade) and 15 repetitions of each treatment, and each plant considered as a repetition. The height and diameter were studied on 60, 82, 103, 124, 145 days after the emergence of the plants. Plants exposed in the shade showed lower vegetation performance than the other conditions tested (50 percent and 100 percent of RFA). The 50 percent luminosity, as well as full sunlight, can be recommended for the formation of seedlings.

ABSTRACT

Objective: To accomplish the detachment and culture of IPE in vitro and to investigate the course of the transdifferentiation from iris pigmented epithelial cell to lens epithelial cell during the specific condition.Methods: The IPE of rabbits were detached and cultivated according to the method of enzyme-assisted microdissection.Then the IPE were cultivated in the culture medium appending with PTU? Huase? rhFGF-4? Asc-2P and so on.Finally the initiative and the ultimate cells were respectively identified by microscope?electroscope and the method of immunohistochemistry.Results: The IPE of rabbits were successfully detached and cultivated and the course of the transdifferentiation was observed.The IPE and LEC were identified by their particular reaction to immunohistochemistry.Conclusion: It is a good method to detach IPE by enzyme-assisted microdissection.During in vitro culture,IPE can undergo the course of transdifferentiation and turn into LEC finally.This experiment suggest that those factors such as PTU? Huase? rhFGF-4? Asc-2P and so on may be involved the regulation of the course of transdifferentiation.
